 I use the extension at the site www.xxxx.gr and the speed of the site is very important to me.

1. I have joomla default sef disabled according the documentation.
But if try to disable virtuemart sef i get 500 error and site won't open until i enable back virtuemart seo. Is this normall?

2. I use jotcache to optimize site speed and a the same time i have configured "shlib - Db query cache and programming library" plugin to work with memcached.
Would it be better to have "Enable db query cache" enabled or not?

The documentation says the exact opposite. You even get a warning in sh404SEF control panel if it's disabled (since version 4.7.0).

But if try to disable virtuemart sef i get 500 error and site won't open until i enable back virtuemart seo. Is this normall?
I'm not so familiar with VM, but I think I know this setting must be on, otherwise VM creates really weird URLs.

If you have an error 500, you need to set Joomla "Error reporting" setting to "maximum" so that more error messages are displayed and you can understand which of the extensions on your site have a problem.

Would it be better to have "Enable db query cache" enabled or not?
I don't know. This setting uses shared memory to cache database query. On some sites, it can be faster, on some it can be slower (because it takes time to store data in cache, and so it's a waste if those data are not reused) or cause issues with some other extensions. You would need to conduct extensive testing to see if it provides any benefit on your site and on your server, there's no one size fits all answer to this question.
