Hi,
When a user click on their avatar or "my Profile" tab, the URL is site.com/go-to-profile

I want the URL for when users go to their profile, is site.com/username

It is possible to do this?

As far as I know, this is not possible. Not because of sh404SEF but because that's just how Jomsocial (and Community builders and other) work: the original, non-sef link doesn't have the user id (which is needed to find the user name).
Jomsocial just creates a (my-profile link), which is the same for all users, and then on that page it displays the profile of the currently logged in user.

That doesn't mean such a URL doesn't exist, but simply on the avatar or the profile page, they don't use it.
